【摘要】 甘油醛-3-磷酸脫氫酶(glyceraldehyde-3-phosphate dehydrogenase,GAPDH)是糖酵解和卡爾文循環(huán)中的關(guān)鍵酶,本實(shí)驗(yàn)室之前的研究發(fā)現(xiàn)斑玉蕈菌絲的GAPDH表達(dá)隨培養(yǎng)基中葡萄糖添加濃度的變化而變化,在此基礎(chǔ)上我們首次克隆了斑玉蕈GAPDH的DNA和cDNA序列,結(jié)果斑玉蕈gpd基因長(zhǎng)2 724 bp,對(duì)比基因組DNA和cDNA序列知其中有7個(gè)內(nèi)含子、8個(gè)外顯子;其理論分子量為36.15 kD、編碼蛋白質(zhì)含338個(gè)氨基酸、等電點(diǎn)(PI)為8.24。
【Abstract】 A glyceraldehyde-3-phosphate dehydrogenase gene(gpd) and corresponding cDNA were cloned from the edible fungus,Hypsizigus marmoreus.Comparison of the cloned DNA and cDNA sequence revealed that gpd was 2 724 bp in length,containing seven introns and eight exons,and encoding a protein consisting of 338 amino acids residues.The calculated molecular weight and PI of the protein were 36.15 kD and pH 8.24,respectively.
