【摘要】 應(yīng)用RAPD技術(shù)對43個賀蘭山紫蘑菇(Cortinarius)屬菌株進(jìn)行遺傳多樣性分析。結(jié)果表明,在供試的17條RAPD隨機(jī)引物中,有7條可擴(kuò)增出清晰、穩(wěn)定的DNA條帶,利用篩選出的7條引物進(jìn)行RAPD擴(kuò)增,共得到79條清晰的DNA條帶,其中68條多態(tài)性片段占總擴(kuò)增片段的86.1%;聚類分析表明,在相似系數(shù)0.63水平時,供試的43個菌株被分為紫紅絲膜菌(C.rufo-olivaceus)和藍(lán)絲膜菌((C.caerulescens)兩個組群,同時紫紅絲膜菌種內(nèi)41個樣品間的差異較顯著,其中種內(nèi)最大相似系數(shù)為0.956,最小相似系數(shù)為0.515;在相似性系數(shù)為0.68時,可將紫紅絲膜菌的41個樣品分為7類。研究結(jié)果表明RAPD技術(shù)可以有效地區(qū)分紫蘑菇菌株并分析種內(nèi)的遺傳多樣性。
【Abstract】 Genetic diversity among 43 strains of Cortinarius was analyzed by RAPD using seven primers selected from 17 test primers previously shown to display specific and stable amplification patterns.A total of 79 RAPD bands were detected,of which 68(86.1%) were polymorphic.A dendrogram was constructed using the amplification data that separated the strains into two groups at a coefficient value of 0.63.The two groups consisted of 41 C.rufo-olivaceus strains and two of C.caerulescens.The 41 C.rufo-oliva...
