【摘要】 建立1種運(yùn)用高效液相色譜(HPLC)測定香菇(Lentinula edodes)中香菇嘌呤含量的方法。優(yōu)化后的色譜條件為:Ultimate AQ-C18柱(5μm,4.6 mm×250 mm),流動相為甲醇-磷酸鹽緩沖液(7∶93,pH 4.67)等度洗脫,流速1 mL/min,柱溫30℃,檢測波長259 nm,進(jìn)樣量10μL。研究結(jié)果發(fā)現(xiàn)該方法準(zhǔn)確、靈敏、重現(xiàn)性好,適用于香菇嘌呤的定量分析。
【Abstract】 A simple and sensitive HPLC-based method for the accurate determination of eritadenine in Lentinula edodes fruit bodies is described.Optimization of sample work-up procedures showed that the efficiency of eritadenine extraction was highest when powdered fruit bodies were suspended in 5%(w/v) ethanol and subjected to ultrasound for 30 min using a 1∶100 sample-solvent ratio.Optimal separation of eritadenine in the extracts was achieved by loading 10 μL samples on to an Ultimate AQ-C18 column(5 μm,...
