【摘要】 研究了云芝子實體氯仿提取物(CVFC)對人肝癌細胞株HepG2的體外抑制作用,并初步探討了抑瘤機制。MTT結(jié)果顯示,CVFC對HepG2細胞有明顯的抑制作用,并呈明顯的量效關(guān)系,半數(shù)抑制濃度為(75.83±3.96)μg.mL-1。熒光顯微鏡和透射電鏡形態(tài)觀察發(fā)現(xiàn)CVFC處理可使細胞發(fā)生細胞皺縮、核碎裂等凋亡特征;Annexin V/PI雙染結(jié)果證實有胞膜磷脂酰絲氨酸的翻轉(zhuǎn),經(jīng)150μg.mL-1 CVFC處理HepG2細胞24 h后早期凋亡率為17.82%,晚期凋亡率為22.59%,均顯著高于對照組5.35%和3.75%。這些結(jié)果表明CVFC具有體外抑制HepG2細胞的活性,其抑瘤機制與誘導(dǎo)腫瘤細胞凋亡有關(guān)。
【Abstract】 In this report,in vitro the inhibition and cytotoxic effects of a chloroform extract from Trametes versicolor fruiting body(CVFC) was studied using a liver carcinoma HepG2 cell line.MTT assay results showed the CVFC inhibited HepG2 cells growth in a dose-dependent manner.The IC50 was found as(75.83±3.96) μg·mL-1.Cell morphological observation using fluorescent microscope and transmission electron microscope revealed that the CVFC treated cells presented typical apoptotic characteristics,includin...
