【摘要】 為確定樺褐孔菌發(fā)酵液提取物的毒性級別,研究對H22肝癌小鼠的抗腫瘤及免疫調(diào)節(jié)作用。依據(jù)2004年實施的《急性毒性試驗國家標(biāo)準(zhǔn)》進行急性毒性研究和級別鑒定,同時建立H22肝癌的荷瘤小鼠模型進行小鼠體內(nèi)抗腫瘤的研究,并采用MTT法檢測荷瘤小鼠自然殺傷細(xì)胞活性(NK)及T淋巴細(xì)胞增殖情況。結(jié)果表明,樺褐孔菌發(fā)酵液提取物(IOFE)的LD50為15.02 g·kg-1>15 g·kg-1,屬無毒級別。與陰性對照和陽性對照比,體重、采食量均穩(wěn)定增長,抑瘤率為44.25%,可顯著提高NK細(xì)胞活性67.13%(p<0.01),但對T淋巴細(xì)胞增殖率效果不明顯。樺褐孔菌發(fā)酵液提取物具有較好的抗腫瘤作用,且毒性極低,抗腫瘤作用可能是通過提高機體免疫活性尤其是NK細(xì)胞的殺傷活性實現(xiàn)的。
【Abstract】 In this work,the toxicity level of the extract from fermention liquid of Inonotus obliquus(IOFE) was tested and the inhibition of hepatoma H22 and immunomodulatory effects in vivo were assessed.According to the acute toxicity tests national standard implemented in 2004,the toxicity level were tested and analyzed.Meanwhile,the models of hepatoma H22 established in mouse were used to study the antiumor effect of IOFE.Then the nature killer(NK) cell acticity and T lymphocyte proliferation rate were detected by MTT method.The results showed that IOFE was no toxicities because LD50 was 15.02 g·kg-1>15.00 g·kg-1.Compared with positive and nagative controls,the body weight and the food intake of mice treated by IOFE increased steadily and the nature killer(NK) cells activity was improved significantly which was 67.13%(p<0.01).Also the inhibition of hepatoma H22 was 44.25%.While the effect on T lymphocyte proliferation rate was not obvious.In conclusion,the tumor can be inhibited well in...更多 vivo by IOFE,and the reason may be that it can imporve the body’s immunity,especially enhance NK cell acticity. 還原
