【中文摘要】 采用熱水浸提法,以粗多糖得率作為評價指標,通過單因素試驗和正交試驗,對花臉香蘑[Lepista sordida(Fr.)Sing.]菌絲體發(fā)酵全液中多糖的提取條件進行優(yōu)化。結果表明,優(yōu)化的提取條件為:提取溫度100℃、提取次數(shù)3次、pH 12、乙醇終濃度70%;在此優(yōu)化條件下,花臉香蘑發(fā)酵全液的粗多糖得率達4 mg/mL。
【英文摘要】 Orthogonal testing [L9(34)] was used to define more precisely the factors,determined on the basis of single factor experiments,affecting crude polysaccharide extraction from submerged cultures of Lepista sordida.Data revealed that the optimal extraction conditions consisted of three sequential aqueous extractions conducted at pH 12 and 100 ℃ for 1 h,followed by precipitation with ethanol at a final concentration of 70%.Under these conditions,the yield of crude polysaccharide extracted was 4 mg/mL.
