【中文摘要】 采用變性梯度凝膠電泳(Denaturing Gradient Gel Electrophoresis)技術(shù)以土壤微生物群體的基因組DNA為研究對象,通過比較羊肚菌(Morchella sp.)出菇土壤及對照土壤中各種真菌的28S rDNA基因信息來了解土壤微生物的多樣性。結(jié)果發(fā)現(xiàn),測得的12種真菌中,盤菌綱的有10種,異擔(dān)子菌綱1種,還有1種未確定。所取2份土樣中僅羊肚菌生長土壤中檢測出有羊肚菌;2份樣品在優(yōu)勢真菌種群上有一定的區(qū)別,盤菌綱真菌群落存在很大的差異,羊肚菌出菇土壤的真菌豐度低于對照,可能與土壤中羊肚菌優(yōu)勢種群對其它種群的抑制作用有關(guān)。
【英文摘要】 Fungal communities inhabiting Morchella growth soil (DM) and common vegetable garden soil (DK) were monitored using Denaturing Gradient Gel Electrophoresis (DGGE). Sequence analysis of 12 recovered dominant bands revealed the presence of ten fungi assigned to the Ascomycota, Pezizomycotina, one assigned to the Basidiomycotina, Heterobasidiomycetes, and one unidentified fungus. Comparison of the dominant fungal groups present in DM and DK revealed large differences in the community composition with respect to Pezizomycete fungi. Fungal communities associated with DM soil were much less varied in composition compared with those associated with DK soil, possibly due to inhibitory effects exerted by the dominant Morchella.
