【中文摘要】 利用RAPD及SRAP分子標(biāo)記技術(shù)對從國內(nèi)外收集到的12個真姬菇菌株進(jìn)行遺傳多樣性分析。結(jié)果表明,20條RAPD引物共擴(kuò)增出多態(tài)性條帶285條,18對SRAP引物共擴(kuò)增出多態(tài)性條帶176條。在相似系數(shù)0.800水平時,RAPD和SRAP分子標(biāo)記分別將12個真姬菇菌株分為5個和6個類群。兩種方法均適用于真姬菇種內(nèi)鑒定,而SRAP標(biāo)記較RAPD標(biāo)記穩(wěn)定性好,更適于真姬菇的種內(nèi)鑒定。
【英文摘要】 Randomly amplified polymorphic DNA(RAPD)and sequence-related amplified polymorphisms(SRAP)were used to identify 12 strains of Hypsizygus marmoreus Sing.A total of 285 and 176 polymorphic bands were amplified by 20 RAPD primers and 18 SRAP primer pairs,respectively.Two dendrograms based on UPGMA(unweighted pair group method with arithmetic average)cluster analysis using the amplification data revealed that,at the similarity level of 0.8,the 12 strains clustered into five and six groups,respectively.We conclude that both methods are suitable for discriminating among strains of H.marmoreus,and that the SRAP markers were more effective in reflecting genetic diversity.
