【中文摘要】 比較香菇(Lentinula edodes)和黑木耳(Auricularia auricula)在不同水解條件下的單位質(zhì)量吸光值.篩選適宜水解條件,并通過測定姬松茸(Agaricus blazei)、雞腿菇(Coprinus comatus)和茶樹菇(Agrocybe aegerita)3種食用菌的總糖含量,對篩選的水解條件進(jìn)行驗證;采用正交試驗優(yōu)化苯酚-硫酸法的反應(yīng)條件;最后選用苯酚硫酸法和費(fèi)林氏滴定法分別測定4種供試食用菌的總糖含量,并進(jìn)行比較。試驗研究表明。食用菌適宜水解條件為:30 mL鹽酸,100℃水浴水解3 h;優(yōu)化的苯酚-硫酸法反應(yīng)條件為:5%苯酚1.5 mL.濃硫酸5.5 mL,反應(yīng)時間15 min;苯酚硫酸法和費(fèi)林氏滴定法兩種方法測得的食用菌總糖含量無顯著差異.因苯酚-硫酸法操作簡單,且簡化了前處理過程.故是國標(biāo)法——費(fèi)林氏滴定法較理想的替代方法之一。
【英文摘要】 Optimal conditions for the hydrolysis of Lentinula edodes and Auricularia auricula fruit body tissue and assay of the total sugar in the hydrolysates using the phenol-sulfuric acid method were determined using orthogonal testing.Optimal hydrolysis was achieved by adding 30 mL conc.HCI to 0.25 g tissue suspended in 100 mL distilled water and incubating at 100℃in a water bath for 3 h.Optimal conditions for the phenol- sulfuric acid assay were:1.5 mL 5% (w/v) phenol,5.5 mL concentrated sulfuric acid,and 15 rain reaction time.These optimized hydrolysis and assay conditions were confirmed by determining the total sugar contcnt of fruit body tissue taken from Agaricus blazei,Coprinus comatus and Agrocybe aegerita.Comparison of the phenol-sulfuric acid and Fehling's methods for total sugar determination of L.edodes,A.auricula,A. blazei and A.aegerita revealed that the phenol-sulfuric acid method was an ideal substitute for the Fehling- titration method.
