【中文摘要】 以獲得弱木栓質(zhì)化靈芝菌株為目的 ,利用液體培養(yǎng) 4d的靈芝菌絲體和培養(yǎng) 3d的糙皮側(cè)耳菌絲體為材料 ,在PEG Ca2 +系統(tǒng)中誘導(dǎo)原生質(zhì)體融合 ,獲融合子 19株 ,融合率為 1.9×10 -5。根據(jù)其生物學(xué)特性 ,選取F3、F8兩融合子及兩親本菌株的基因進(jìn)行RAPD分析。結(jié)果表明 ,兩親本的核物質(zhì)在融合子中發(fā)生了重組 ,并影響了酶蛋白的表達(dá) ,而融合子在DNA水平上更接近靈芝
【英文摘要】 In order to obtain less corkification strain of G.lucidum, the mycelia of G.lucidum and P.ostreatus were put under submerged culture for 4d and 3d respectively to prepare the protoplast. The fusion experiment between G. lucidum and P. ostreatus protoplasts was carried out under PEG Ca 2+ system, and nineteen fusants were selected.The fusion frequency was as high as 1.9×10 -5 . According to their biological characters, RAPD analysis for genomes of fusants F3?F8 selected randomly and two parents was conducted. The results showed that the nuclear substance of two parents was recombined in the fusant and the expression of the enzyme protein was affected. The fusants were proved to be more similar to G.lucidum at DNA level.
