【中文摘要】 棘托竹蓀 (DictyophoraechinovolvataZang ,ZhengetHu)子實(shí)體干品經(jīng)熱水抽提 ,乙醇沉淀 ,蛋白酶法和Sevag法去蛋白后得粗多糖DE。DE經(jīng)乙醇分級(jí)沉淀得到三個(gè)級(jí)分DE1、DE2和DE3。DE2經(jīng)DEAE SephadexA - 2 5柱進(jìn)一步純化得到棘托竹蓀多糖純品DE2 - 2。DE2 - 2經(jīng)高效液相色譜法測(cè)定為均一物質(zhì)。紅外掃描表明 ,其具有典型的多糖吸收峰。紙層析和氣相色譜分析結(jié)果表明 ,DE2 - 2的單糖組成為 :D -葡萄糖、D -甘露糖、D -半乳糖和L -巖藻糖 ,其摩爾比為 8.68∶1.0 0∶1.85∶0 .74 ,分子量約為 84 0 0 0。經(jīng)測(cè)定 ,DE2 - 2具有一定的抑瘤活性 ,其抑瘤率為38.93%。
【英文摘要】 The crude polysaccharide DE was extracted from dried fruitbodies of Dictyophora echinovolvata by hot water extraction, ethanol precipitation and deproteinization with protease method and Sevag method.Then three fractions DE1, DE2 and DE3 were obtained by fractional precipitation of DE with ethanol. While pure DE2-2 was obtained by further purification of DE2 with DEAE Sephadex A-25 and its homogeneity was verified by HPLC. And typical absorption peaks of polysaccharide were found with IR-scan. It was proved that DE2-2 was composed of D glucose, D mannose, D galactose and L fucose with a molar ratio of 8.68∶1.00∶1.85∶0.74 by paper and gas phase chromatography. The molecular weight of DE2-2 was estimated to be 84000 with an inhibition rate of 38.93% to Sarcoma-180 implanted in mice.
