【中文摘要】 從我國(guó)使用的 19個(gè)主要香菇栽培菌種中制備了 2 5種原生質(zhì)體單核體 ,以此為材料 ,以交配型基因?yàn)闃?biāo)記 ,在交配反應(yīng)和RAPD兩個(gè)水平上對(duì)這 19個(gè)菌株進(jìn)行了較為詳細(xì)的遺傳相似性分析。 2 5種原生質(zhì)體單核體兩兩配對(duì)共 30 0個(gè)組合 ,其中不親和性反應(yīng)為 110對(duì) ,占配對(duì)總數(shù)的 36 .7%。根據(jù)交配反應(yīng)結(jié)果 ,2 5個(gè)單核體可分為三大類群、7個(gè)交配型組。這些結(jié)果從交配型角度證明 ,這 19個(gè)菌株具有十分狹窄的以交配型基因?yàn)闃?biāo)記的遺傳背景。RAPD反應(yīng)和聚類分析中 ,這三大類群、7個(gè)交配型組基本上都能聚為一類 ,從而在分子水平上驗(yàn)證了交配型分析的結(jié)果 ,也為交配型基因在菌種鑒定中的作用提供了依據(jù)。
【英文摘要】 protoplasted monokaryons were prepared from 19 Lentinula edodes cultivating strains in China. Genetic similarity analysis of these 19 strains was carried out based on the mating reaction and RAPD analysis of 25 monokaryons with mating type genes as marker. It was found that the incompatibility reaction rate was 36.7% in 300 progenies from 25 protoplasted monokaryon pairings. Based on the mating reaction results, 25 protoplasted monokaryons could be divided into three classes with 7 mating type groups. And the results also indicated that these 19 strains had high genetic similarity. Similar results were obtained by RAPD analysis and cluster analysis. This implicated that mating type genes could be used for strain identification.
