【中文摘要】 采用凝膠電泳等生化技術(shù)探討了蘑菇易褐變菌株的酚酶特征。發(fā)現(xiàn)易褐變菌株具有特異的同工酶表型,子實(shí)體階段表達(dá)的酚酶位點(diǎn)多,且酶活性高;其整個(gè)生活史的胞內(nèi)酚酶活性曲線變化與優(yōu)質(zhì)菌株不同;所表達(dá)的酚酶對(duì)亞硫酸鹽類(lèi)酚酶抑制劑的敏感性低;所表達(dá)的酚酶較耐熱,在加熱的初始階段(35~40℃)就能迅速催化褐變反應(yīng);所含的潛伏型酚酶含量大,且隨著鮮菇貯藏期延長(zhǎng)而大量活化,繼續(xù)催化褐變。本文還就如何防止褐變進(jìn)行了探討。
【英文摘要】 The polyphenol oxidase (PPO) characteristics of easy-browning A. bisporus strains were investigated using biochemical technology such as AGE etc. The results showed that the easy-browning strains were possessed of special PPO isozyme pattern and more PPO loci with high enzyme activity at fruitbody growing stage, that the changes of the endocellular PPO activity curve of easy-browning strains were different from that of the high-quality strains during its whole life cycle, that its PPO sensitivity to the PPO inhibitor such as Na2SO3 was relatively low, that its PPO which was tolerant to heat could swiftly catalyze the browning reaction during the initial stage of heat-ing(35-40℃) and that its content of latent PPO was great and the latent PPO was activated in great amount with the extending of storage time to continue catalyzing the browning reaction. And the control of the browning was also discussed in this paper.
