【中文摘要】 通過研究培養(yǎng)時(shí)間和培養(yǎng)基種類對毛木耳同工酶凝膠電泳圖譜的影響,證實(shí)酯酶同工酶在毛木耳中是一種穩(wěn)定表達(dá)的酶類,培養(yǎng)時(shí)間和培養(yǎng)基種類對其無明顯影響;蘋果酸脫氫酶同工酶不受培養(yǎng)基種類的影響,但隨培養(yǎng)時(shí)間的延長,由5~10d時(shí)的三條帶減為15~25d時(shí)的二條帶;多酚氧化酶同工酶和過氧化物酶同工酶受培養(yǎng)叢種類的影響顯著,但除培養(yǎng)初期酶帶數(shù)較少外,在10~25d時(shí)酶帶數(shù)穩(wěn)定不變。由此可篩選最適培養(yǎng)時(shí)間和培養(yǎng)基種類,用于木耳屬菌株的分類鑒定。
【英文摘要】 Effects of different culture time and medium on the isozyme gel electrophoretic pattern of Auricu-laria polytricha were investigated in this paper. The results showed that the esterase isozyme was a stable expressing enzyme in A . polytricha, its pattern was not significantly affected by the culture time and medium.The malate de-hydrogenase isozyme was not affected by the medium, but with the elongating of the culture time, the band numbers were decreased from 3 in 5- l0d to 2 in 15-25d. The polyphenol oxidase isozyme and peroxidase isozyme were sig-nificantly affected by different media, but the band numbers were stable in 10-25d except that the band numbers were less in early culture time of 5d. These results indicated that we could choose optimal culture time and medium to classify and identify the strains in Auricularia Bull. ex Mer.
