【中文摘要】 金針菇與鳳尾菇皆屬四極性異宗結(jié)合食用菌,其雙核異核體菌株都具鎖狀聯(lián)合遺傳標(biāo)記。本研究以金針菇和鳳尾菇的雙核異核菌株為親本,將熱滅活的鳳尾菇原生質(zhì)體,以PEG為融合劑,在高鈣、高pH值條件下,與金針菇原生質(zhì)體融合,結(jié)果從1329個(gè)再生菌株中選擇出6株雙親細(xì)胞質(zhì)和細(xì)胞核都融合的無(wú)鎖狀聯(lián)合單核菌株,經(jīng)融合核分裂技術(shù)處理后,融合核分裂成為具鎖狀聯(lián)合的雙核菌株。應(yīng)用RAPD技術(shù)分析,證明融合子菌株均含有與雙親株同源的遺傳物質(zhì),是真正的融合子菌株。
【英文摘要】 Flammulina velutipes (Fr. ) Singer and Pleurotus sajor-caju (Fr.) Singer were tetrapolar and heterothallic edible fungi and their hetero-dicaryotic strains, selected as parent strains in this study, were both with clamp connection as genetic marker. F. velutipes protoplast and heat-treated P. sajor-caju protoplast were fused under high concentration of Ca2+and high pH conditions using PEG as fusogen. And 6 uninucleate fusants without clamp connection, with cytoplasm and cell nuclei of parent strains being all fused, were identified from 1329 regenerate strains. Using fused nucleus division technique, the fused nucleus was divided into dikaryotic strain with clamp connection. While fusants containing the same genetic material as the parent strains was proved to be the genuine fusants by RAPD technique.
