【中文摘要】 利用等高鎖狀同源電場(chǎng)凝膠電泳(CHEF),獲得草菇的電泳核型.綜合兩組脈沖電泳的電泳條件、溴化乙錠染色以及DNA雜交分析結(jié)果,草菇染色體DNA至少可分為9個(gè)譜帶.這一結(jié)果和以前用光學(xué)顯微鏡觀察草菇的染色體數(shù)目的結(jié)果相一致.以混濁釀酒酵母和粟酒裂殖酵母等的相對(duì)遷移率為基準(zhǔn),草菇染色體長(zhǎng)度估計(jì)在1.32~5.4Mb之間,染色體組總長(zhǎng)約25Mb.
【英文摘要】 An electrophoretic karyotype of Volvariella volvacea was obtained using Contour-Clamped Homogeneous Electric Field (CHEF) gel electrophoresis. Based on the conditions of 2 sets of pulsed field gel electrophoresis, the results of ethidium bromide staining and DNA hybridization analysis with two moderately repetitive probes, it was concluded that at least 9 chromosomal DNAs were found in V. volvacea. This result was consistent with previous studies using light microscopy. The chromosome size was estimated as 1.32 Mb to 5.4 Mb based on gel migration relative to the chromosomes of Saccharomyces cerevisiae, Schizosaccharomyces pombe and Hansenula wingei strains. The total genome size of V. volvacea was estimated to be about 25 Mb.
