【中文摘要】 側(cè)耳、香菇皆屬四極性異宗結(jié)合食用菌,其異核雙核菌絲體均具“鎖狀聯(lián)合”形態(tài)標(biāo)記。本研究以PEG-Ca++為誘導(dǎo)劑,以紫孢側(cè)耳和香菇Cr-20的原生質(zhì)體再生單核體為親株,進(jìn)行側(cè)耳與香菇的屬間原生質(zhì)體融合,結(jié)果從1022個融合再生體中鏡檢出16個具鎖狀聯(lián)合遺傳標(biāo)記的融合株。對1~4號融合株與親株進(jìn)行的拮抗反應(yīng)、雙單交配、過氧化物同功酶譜分析及菇形特征對比皆證明融合株確具雙親雜合體特征。出菇驗證表明,融合株菌絲體長速均快于親株,出菇期早,菇產(chǎn)量近于或高于親株側(cè)耳的水平。
【英文摘要】 Pleurotus sapidus and Lentinus edodes are tetrapolarity heterothallic edible fungus, the heterokaryotic dikaryotic mycelia had morphology marker of clamp conncnction (C. C. ). In this study, intergeneric protoplast fusion was carried out using regenerative uninuclear protoplasts from P. sapidus and L. edodes Cr-20 as parent strains, using PEG-Ca++ as inducer. 16 fusion strains with genetic marker of C. C. were obtained from 1022 regenerative strains. Antagonistic experiment, di-mon mating, peroxidase isoenzyme pattern anylysis, comparison of fruit body morphology and characters were conducted for 4 fusion strains. The test showed that the fusion strains have hybrid characters of the parents. In addition, the yeild of the fusion strains, with mycelia growth rate faster and fruiting period earlier than that of their parents, was comparable to or higher than that of Pleurotus parent.
