【中文摘要】 用60Co-γ射線輻射誘變佛羅里達(dá)側(cè)耳孢子,得到抗NaN_3藥物的突變體。突變體的原生質(zhì)體經(jīng)0.83%碘乙酰胺滅活處理,與姬菇單核菌絲釋放的原生質(zhì)體進(jìn)行融合,然后涂布在1×10-6NaN_3藥物選擇再生培養(yǎng)基上,得到一個融合子菌落。經(jīng)酯酶同工酶分析表明:融合子的12條區(qū)帶中,與雙親共有的區(qū)帶5條,與佛羅里達(dá)側(cè)耳親本同者2條,與姬菇親本同者3條,新菌株獨具的區(qū)帶2條,證實為雙親融合雜種。
【英文摘要】 The antidrug NaN3 mutants were obtained by radiating the spores of Pleurotus florida with 60Co. After being treated by 0.83% CH2 ICONH2 , the protoplasts of mutants were mixed with protoplasts released by uninucleate mycelium of Pleurotus sp. Then spreading it on a selected-regeneration-medium containing 1×10-6NaN3, a fusant colony was obtained at last. Analysis with esterase isozymes zymograms proved that among the twelve bands of fusants, five shared the same bands with two parents, two were the same to that of Pleurotus florida, three were the same to that of Pleurotus sp. and two were new bands. So it was identified that the new hybrid we obtained was fusant hybrid of two parents.
