【中文摘要】 為了提高蛹草擬青霉cmily-02產(chǎn)蟲(chóng)草素得率,采用正交實(shí)驗(yàn)優(yōu)化固體發(fā)酵培養(yǎng)基配方。以蟲(chóng)草素產(chǎn)量為指標(biāo),通過(guò)高效液相色譜法檢測(cè),篩選出最優(yōu)固體培養(yǎng)基為:大米粉2.0%、青稞粉0.5%、玉米粉3.0%、黃豆粉0.5%、粗麥麩30%、KH2PO3 0.05%、MgSO4 0.1%。從培養(yǎng)溫度和干燥溫度2方面,探討了溫度對(duì)蟲(chóng)草素產(chǎn)量的影響,得出產(chǎn)蟲(chóng)草素的最佳培養(yǎng)溫度為28℃,最佳干燥溫度為30℃。通過(guò)固體發(fā)酵條件優(yōu)化,蟲(chóng)草素產(chǎn)量可達(dá)到1 850μg.g-1。
【英文摘要】 In order to increase the yield of cordycepin in the culture mycelia of Paecilomyces militaris cmily-02,the best solid culture medium was screened by orthogonal tests and the content of cordycepin in the culture mycelia was determined by HPLC.The results showed that the optimum medium components were rice flour 2.0%,highland barley flour 0.5%,indian meal 3.0%,soybeans meal 0.5%,outerbran 30%,KH2PO4 0.05%,MgSO4.7H2O 0.1% and so on.The optimum culture conditions were culture temperature 28℃,drying temperature 30℃ with the total yield of cordycepin reaching as high as 1 850 μg.g-1.
