【中文摘要】 目的:在前期藥理實驗基礎(chǔ)上,分析雙向性固體發(fā)酵后雷公藤菌質(zhì)的化學(xué)成分。方法:采用高效液相色譜法和薄層色譜法,研究雷公藤藥材和不同發(fā)酵時間藥性菌質(zhì)的化學(xué)成分動態(tài)變化規(guī)律。結(jié)果:藥性菌質(zhì)的雷公藤甲素含量低于原藥材低;分別在218nm和250nm波長下,高溫滅菌藥材和發(fā)酵30d的藥性菌質(zhì)的液相圖譜有3處明顯的變化;乙酸乙酯提取物的薄層層析發(fā)現(xiàn)有一藍色斑點為發(fā)酵后新增成分,而甲醇提取物薄層層析發(fā)現(xiàn)有2個斑點隨發(fā)酵時間的延長含量降低。結(jié)論:雷公藤經(jīng)藥用真菌發(fā)酵后化學(xué)成分發(fā)生變化,其中一些化學(xué)成分的變化規(guī)律與藥理實驗結(jié)果存在相關(guān)性,可作為發(fā)酵終點判斷標準。
【英文摘要】 Objective:On the base of previous pharmacological and toxicological study,chemical compositions in Ttripterygium wilfordii HooK.F after fermentation was compared in this paper.Methods:The content of triptolide in each medicinal material was determined with HPLC and chemical constituents was analyzed with HPLC and TLC.Results:Triptolide content decreased after raw plant herbs was fermented with medicinal fungi,there were three characteristic absorption peaks at wavelength of 218nm and 250nm,one blue spot in TLC chromatograms of ethyl acetate extract and two spots gradually disappeared with the fermentation extent.Conclusion:The of chemical constituents in Tripterygium wilfordii HooK.F happened when medicinal material was fermented,there was relativity of chemical compositions and pharmacological toxicology,consequently,it was regarded as one standard of fermentation termination.
