【中文摘要】 概述了食用菌轉(zhuǎn)化取得的進(jìn)展和存在的問題,詳細(xì)介紹了篩選標(biāo)記、啟動子、轉(zhuǎn)化方法、整合方式以及將來的研究方向等方面的研究狀況。篩選標(biāo)記主要有營養(yǎng)缺陷型、藥物抗性標(biāo)記;啟動子主要是同源啟動子ras基因和gpd基因啟動子;轉(zhuǎn)化方法主要有PEG法、電擊法、限制性內(nèi)切酶介導(dǎo)法、農(nóng)桿菌介導(dǎo)法,對比分析了各種方法的優(yōu)缺點;重組方式主要是多位點整合;甲基化是導(dǎo)致轉(zhuǎn)錄水平低的一個原因;可能的提高轉(zhuǎn)化率措施有采用強啟動子、加入同源片段進(jìn)行同源整合。
【英文摘要】 The progress and the problems in the transformation of edible fungi were summarized after introducing the select marker,the promoter,the ways of transformation,the pattern of integration and the future direction of the research in detail.Two main select markers were auxotroph and drug resistance.The promoter were major the ras gene of homologous promoter and the gpd gene promoter;the methods of transformation were main PEG method,electroporation method,restrictive enzyme-mediatedintegration and Agrobacterium-mediated method,which were advantages and disadvantages respectively after compared analyzing.The style of recombination is integration on multiple locus.The heavy methylation of the integrated DNA was one reason of low transcript.The possible measures which improved the frequency of transformation made homologous recombination by using strong promoter and adding homologous sequence.
