【中文摘要】 本研究以RAPD(RandomlyAmplifiedPoly-morphicDNA)技術(shù)為主,比較分析食用菌木耳屬(Auricularia)種內(nèi)雜交種.種間原生質(zhì)體融合子與其親本菌株的PCR(PolymerseChaihReaction)擴增的DNA片段之間差異,并輔以其它手段,如核相及核數(shù)目觀察,同功酶鑒別,以期建立完整有效的檢測技術(shù)系列,確證其雜交種融合子的真實性及可靠性。在RAPD分析中,用于測試的10種隨機引物僅有4種引物與供試菌株的DNA模板具有較好的結(jié)合能力,其中1種引物可用于檢測雜交種及其親本,另外3種引物可用于檢測融合子反親本。檢測結(jié)果顯示:3個供試雜交種中有2個為真雜合子,1個為假雜合子;2個供試融合子均為非穩(wěn)定雜合子。與常規(guī)方法鑒定結(jié)果比較基本相符。因此,RAPD技術(shù)提供了一種快速、簡便、準(zhǔn)確的雜交種及融合子的鑒別方法。
【英文摘要】 RAPD(Randomy Amplified Polymorphic DNA)was used to analysethe differences of DNA fragments between intraspecies hybrid,interspecies protoplastfusants and their respective parents in genus Auricularia,With the help of other methodssuch as the nuclear number in the hyphal cell, the comparison of the isozyme graphs, the hybrids and the protoplast fusants were identified.Four of ten arbitrary primets had good compatibility with DNA template of test strainsin RAPD assay. One of four was used to dentify by brids and their parents, three of fourwere used to identify fusants and their parents. The results showed that two of threehybrids were real heterozygotes,the two fusants were not stable.Therefore,comparing with the other ways,RAPD provided a rapid, simple, accurateidentified method of hybrids and fusants.
