【中文摘要】 從桃紅平菇單核菌絲分離出原生質(zhì)體鋪于含0.6mol/l蔗糖的CM表面,經(jīng)紫外線處理一定時(shí)間后,培養(yǎng)7—10天后長(zhǎng)出的菌落被轉(zhuǎn)于MM之上,在其上生長(zhǎng)十分緩慢或停止的菌落通過(guò)各種營(yíng)養(yǎng)添加物測(cè)其營(yíng)養(yǎng)需求,最后得到一個(gè)需要補(bǔ)充丙氨酸和天冬氨酸才能正常生長(zhǎng)的雙缺突變體。試驗(yàn)表明,該突變體能很好地分離出原生質(zhì)體,且在含0.6mol/l蔗糖的MM上不能再生,因而具有較好的應(yīng)用前景。
【英文摘要】 Protoplasts isolated from monokaryotic mycelia of Pleurotus salmonea-straminens were spread on CM containing 0.6 mol/l sucrose and irradiated by U. V. After 7—10 days, the regenerated colonies were transplanted on MM and those showing little growth were detected further with various kind of nutritional requirement., One auxotrophic mutant, double deficient in Ala and Asp, was obtained at last; from which a good protoplast yield could be obtained. It will be useful in protoplast fusion study because its protoplasts can not regenerate on MM containing 0.6 mol/l sucrose.
