【中文摘要】 本文報(bào)導(dǎo)使用Novozym234酶從光木耳(Auricularia auricula)和毛木耳(Auricularia polytricha)單核菌絲體中,成功地分離出高產(chǎn)量的原生質(zhì)體;一些因素對(duì)原生質(zhì)體形成的影響及原生質(zhì)體再生過(guò)程中形態(tài)學(xué)變化的觀察結(jié)果。菌體的液體培養(yǎng)時(shí)間、緩沖液pH值和一定濃度的酶液用量與原生質(zhì)體的形成有著密切的關(guān)系。原生質(zhì)體的兩種再生型能在液體的CYM(Complete yeast extract medium)和固體的CYM含有0.6M山梨醇的再生培養(yǎng)基中觀察到。
【英文摘要】 The formation and regeneration of mycelial protoplasts from Auricularia auricula and Auricularia polytricha using Novozym 234 were examined. The amour of release of protoplasts is related to mycelial age, buffet pH and dose of enzyme used. In general, liquid culture for 10 days or buffer pH 5—5.9 and the 3ml 2% of Novozym 234 per gram wet weight mycelia are suitable for protoplast formation in both strains. The regeneration frequency of protoplasts of both strains was found by using RM medium, 3.4% for Auricularia polytricha and 3.2% for Auricularia auricula. Tworvegeneration types were observed: Protoplasts directly produced one or more germ tubes and developed into bud-like structures of cells before producing germ tubes by using LRM medium.
