【摘要】 采用FIASCO方法,從糙皮側(cè)耳(Pleurotus ostreatus)菌株ACCC 50838分離簡(jiǎn)單重復(fù)序列分子標(biāo)記(Simple sequence repeat,SSR),所得標(biāo)記用于分析我國(guó)52個(gè)側(cè)耳(平菇)栽培菌株多樣性,對(duì)于SSR標(biāo)記相同的菌株再進(jìn)行ISSR PCR分析。結(jié)果表明:18個(gè)SSR標(biāo)記在供試菌株中的等位基因的數(shù)量為3～12個(gè),多態(tài)性指數(shù)(PIC)為0.34～0.84;除了2組(ACCC50618,ACCC 50866和ACCC 50915;ACCC 50249和ACCC50476)共5個(gè)菌株之外,其他47個(gè)菌株都能利用這18個(gè)SSR標(biāo)記進(jìn)行有效的鑒別。SSR標(biāo)記相同的的菌株,其ISSR、體細(xì)胞不親合性和出菇性狀(溫型、菌蓋顏色)也相同,這些菌株可能是同物異名。利用這18個(gè)SSR標(biāo)記分析52個(gè)菌株的遺傳多樣性,相似性指數(shù)為0.755～0.9995。根據(jù)相似性指數(shù)進(jìn)行聚類分析,52個(gè)菌株分成兩個(gè)大的類群,分別為糙皮側(cè)耳(P.ostreatus)和肺形側(cè)耳(P.pulmonarius)。結(jié)合這些栽培菌株的農(nóng)藝性狀和SSR分子標(biāo)記的聚類分析結(jié)果,我國(guó)平菇栽培菌株的遺傳...
