響應(yīng)面法優(yōu)化靈芝AM21菌株液體深層發(fā)酵培養(yǎng)基配方

【摘要】 為了提高靈芝(Ganoderma lucidum)AM21菌株菌絲產(chǎn)靈芝三萜的能力,采用響應(yīng)面法對其液體發(fā)酵培養(yǎng)基配方進行優(yōu)化。首先采用Plackett-Burman設(shè)計法對影響菌絲體三萜含量的9個相關(guān)因素進行篩選,確定主要影響因子為豆粕粉和山藥,然后用最陡爬坡試驗逼近2個主要因素的最大響應(yīng)區(qū)域,并通過中心組合設(shè)計和響應(yīng)面分析,確定主要影響因子的最佳濃度。優(yōu)化后的培養(yǎng)基組成為1.84%豆粕粉,1%玉米粉,1%葡萄糖,0.92%山藥,0.5%酵母粉,0.1%麩皮,0.1%KH2PO4,0.1%MgSO4.7H2O,0.01%VB1。使用該培養(yǎng)基生產(chǎn)的靈芝菌絲體中三萜含量較使用基礎(chǔ)發(fā)酵培養(yǎng)基生產(chǎn)的高29.13%。
【Abstract】 A fermentation medium for producing mycelium of Ganoderma lucidum,strain AM21,with enhanced triterpenoid content was optimized using response surface methodology.A Plackett-Burman design was applied to screen potential key factors,and the steepest ascent method was adopted to approach the approximate optimal medium composition.Finally,central composite design and response surface methodology were applied to define more precisely the optimal composition of the fermentation medium.Our data reveale...
