樺剝管孔菌液體發(fā)酵環(huán)境條件的研究

【摘要】 以發(fā)酵液的多糖含量為考核指標(biāo),通過單因素試驗(yàn)和正交試驗(yàn),對(duì)樺剝管孔菌(Piptoporus betulinus)液體發(fā)酵的接種量、培養(yǎng)基裝量及培養(yǎng)基初始pH進(jìn)行優(yōu)化。結(jié)果表明,優(yōu)化的樺剝管孔菌培養(yǎng)條件為:接種量2.5%、500mL三角瓶裝200mL液體發(fā)酵培養(yǎng)基、培養(yǎng)基初始pH自然條件下,發(fā)酵6d時(shí),發(fā)酵液中多糖含量最高(28.75%)。
【Abstract】 Orthogonal testing [L9(33)] was used to define more precisely the factors,determined on the basis of single factor experiments,affecting extracellular polysaccharide production in submerged cultures of Piptoporus betulinus.Data revealed that the optimal culture conditions consisted of 2.5% inoculum size,200 mL medium in a 500 mL culture flask,and an initial pH of 5.6.Under these conditions,total polysaccharide concentrations reached 0.18 mg/mL.
