【摘要】 對(duì)吉林省延吉市和云南省昆明市松茸子實(shí)體的ITS序列進(jìn)行遺傳多樣性研究。提取兩個(gè)地區(qū)松茸子實(shí)體的基因組DNA,應(yīng)用特異引物對(duì)其ITS序列進(jìn)行PCR擴(kuò)增及測(cè)序,并對(duì)云南省昆明市松茸樣品進(jìn)行克隆及測(cè)序。之后從GenBank上獲取5個(gè)地區(qū)7條松茸ITS序列,并利用MEGA、BLAST、DNAMAN和ClustalX軟件進(jìn)行對(duì)比分析和構(gòu)建系統(tǒng)發(fā)育樹(shù)。對(duì)云南松茸樣品克隆發(fā)現(xiàn),ITS序列中有A與G、T與C的顛換,并插入1個(gè)堿基T。序列對(duì)比發(fā)現(xiàn),松茸ITS序列長(zhǎng)度在600 bp~604 bp范圍內(nèi),G+C含量在43.0%~44.2%之間,共有41個(gè)變異位點(diǎn),ITS2變異位點(diǎn)數(shù)要多于ITS1的變異位點(diǎn)數(shù)。同時(shí),多個(gè)位點(diǎn)存在著轉(zhuǎn)換、顛換、插入和缺失,表明不同產(chǎn)地松茸ITS序列在進(jìn)化過(guò)程中存在差異,為真菌分類(lèi)、鑒定等研究提供了重要的資料和依據(jù)。
