    摘要:本文對采自湖南莽山的26種鵝膏菌屬(Amanita)真菌進(jìn)行了隨機(jī)擴(kuò)增多態(tài)性DNA (RAPD)分析,40個隨機(jī)引物中篩選出擴(kuò)增效果較好的6個引物,每個引物能產(chǎn)生1~10條DNA條帶,獲得的RAPD譜帶清晰并呈現(xiàn)多態(tài)性O(shè)PG15、OPH04兩個引物擴(kuò)增的RAPD譜帶能將26種鵝膏菌完全區(qū)分開來,通過6個引物的RAPD分析獲得的平均相似性系數(shù)表明種與種之間的相關(guān)系數(shù)在20D60%之間,平均鏈鎖聚類分析可將26種鵝膏菌分為二大類,且一些具菌環(huán)和苞狀菌托的種類聚在一起,與形態(tài)分類基本相吻合。

    STUDY ON RELATIONSHIP AMONG SPECIES OF

    AMANITA USING RAPD

    ABSTRACT: Random amplified polymorphic DNA (RAPD) was used in analysing the relationship among twenty-six speices of Amanita from Mangshan, Hunan. Of 40 primers, 6 ones generated reproducible RAPD profiles, and each one produced 1-10 bands. Two primers (OPG15 and OPH04) could differentiate 26 species of Amanita by its RAPD profiles. The similarity coefficients obtained from profiles generated by six primers among Amanita were about 20-60%. A dendrogram was constructed using average linkage clustering for clustering analysis. It indicated that there were two groups in 26 species and some species with ring and membranous volva clustered a group which coincided with morphological classification.
