黑木耳電泳核型分析

摘要:采用等高鎖狀均質(zhì)電場(chǎng)(CHEF)凝膠電泳技術(shù),對(duì)黑木耳(Auricularia auricula)電泳核型進(jìn)行分析,以釀酒酵母(Saccharomyces cerevisiae)菌株YPH755和粟酒裂殖酵母(Schizosaccharomyces pombe)菌株AS2.214的染色體DNA大小作為分子量標(biāo)記,估計(jì)黑木耳基因組中至少包含9條DNA分子量在850kb至5800kb之間的染色體,多數(shù)染色體DNA分子量在1000kb至3400kb之間,基因組大小在22Mb以上。

ABSTRACT: Electrophoretic karyotype of jelly basidiomycetes Auricularia auricula was analysed by use of contour-clamped homogeneous electric field (CHEF) electrophoresis. Using chromosomal size standards from Schizosaccharomyces pombe AS2.214 and Saccharomyces cerevisiae YPH755, the chromosomal number of A. auricula was estimated to be at least 9 ranging in size from 850kb to 5800kb. The total genome size was over 22Mb.
