草菇耐低溫菌株的誘變選育與鑒定
SELECTION FOR COLD-RESISTENT STRAIN BY COMPLEX MUTAGENESIS OF VOLVARIELLA VOLVACEA AND IDENTIFICATION OF THE STRAIN

摘 要:在草菇V23原生質(zhì)體形成和再生的最佳條件下制備原生質(zhì)體.分別用紫外線(UV),60Co-γ射線,硫酸二乙酯(DES)對原生質(zhì)體進(jìn)行復(fù)合誘變.初篩、復(fù)篩獲得耐常規(guī)低溫(4℃)10d的菌株VH.栽培實(shí)驗(yàn)證明:VH在26℃下的生物學(xué)效率明顯優(yōu)于V23(CK),且差異極顯著.同工酶分析表明:VH過氧化物同工酶,酯酶同工酶譜帶數(shù)量及酶活性均發(fā)生變化;RAPD實(shí)驗(yàn)顯示:VH相對于V23的變異系數(shù)為0.213.VH菇體正常,味道鮮美,食用后無毒副作用.氨基酸組成分析表明:除總氨基酸和谷氨酸含量VH高于V23外,其它氨基酸差異不明顯.VH遺傳性狀穩(wěn)定,是具有潛在應(yīng)用價值的優(yōu)良菌株.
