靈芝菌絲原生質(zhì)體形成與再生的研究

　　摘要：本文探討了靈芝(Ganoderma lucidum)菌絲原生質(zhì)體的制備及再生的條件。實(shí)驗(yàn)結(jié)果表明：采用MYG為靈芝菌絲培養(yǎng)基，2%真菌溶壁酶(Lywallzyme)作脫壁酶，0.6mol／L甘露醇作滲透壓穩(wěn)定劑；對培養(yǎng)6d的靈芝菌絲在30℃下進(jìn)行酶解，酶解時(shí)間4-6h，可產(chǎn)生約3.0×106個(gè)／mL(0.1g菌絲)原生質(zhì)體，并在添加靈芝子實(shí)體浸出汁的雙層培養(yǎng)基上實(shí)現(xiàn)了靈芝菌絲原生質(zhì)體的再生。

Formation　and　Regeneration　of　Protoplast　From

the　Mycelia　of　Ganoderma　lucidum

　　Abstract:The conditions of protoplast formation and regeneration from the mycelia of Ganoderma lucidum were studied.Experimental results indicated that 3.0×106／mL protoplast of the mycelia of Ganoderma lucidum was obtained under optimal conditions,which are described as follows:Using 0.6mol／L mannitol as osmotic stabilizer;2% Lywallzyme as the enzyme digesting the cell wall;Using 6 days old MYG liquid cultures of the mycelia Ganoderma lucidum；at 30℃,enzyme digesting 4～6 hours.When incubated on two layer plates of agar,the protoplasts regenerated and formed new mycelia of Ganoderma lucidum.
