裂褶菌產纖維二糖脫氫酶條件優(yōu)化及
部分酶學性質研究

摘 要:研究了裂褶菌Schizophyllum communeAS 5.391產纖維二糖脫氫酶的條件。該酶是誘導酶,棉花是最好的誘導底物。培養(yǎng)基中加入琥珀酸鈉緩沖液和土溫80利于酶的合成。而木素相關物質藜蘆醇或愈創(chuàng)木酚對酶的生成沒有影響。該酶在pH3.5~10.5和45℃以下保持穩(wěn)定,其最適作用溫度為30℃,最適pH為4.5。Zn2+和Ag+對該酶活性有很大的抑制作用,而疊氮化鈉和氰化鉀對酶活力沒有影響。

OPTIMIZATION OF CELLOBIOSE DEHYDROGENASE
PRODUCTION BY SCHIZOPHYLLUM COMMUNE
AND SOME PROPERTIES OF THE ENZYME

ABSTRACT: Effects of various parameters on cellobiose dehydrogenase (CDH) production by Schizophyllum commune AS 5.391 were determined. CDH was induced by cellulosic substrates, and cotton cellulose with high crystallinity supported the maximum CDH production. Addition of succinate buffer and Tween 80 facilitated CDH secretion. The optimized temperature and pH for CDH are 30℃ and 4.5. It keeps stable at pH range of 3.5~10.5, and below 45℃. CDH was inhibited by Zn2+ and Ag+, but it was not influenced by azide and cyanide.
